Which HLA loci can be sequenced with NGSgo, NGS-Turbo, and NGS-Pronto products?
NGSgo®, NGS-Turbo® and NGS-Pronto® products are available for the following HLA loci: HLA-A, B, C, DRB1, DRB3/4/5, DQA1, DQB1, DPA1, DPB1. Additionally, NGSgo is available for HLA-G (RUO only).
Are there products for non-classical HLA or other genes available?
Yes, we have prototype reagents available for several genes. NGSgo-MoreMX includes HLA-DRA, -E, -F, -G, and -H, and HLA-related genes MR1, MICA, and MICB. Additionally, singleplex prototypes are available for HLA-E and HLA-F. Amplification strategies are also available for HLA-G, KIR and MICA/MICB (RUO). In addition, there is a prototype amplification strategy for 11 rare blood group systems.

What is the difference between NGSgo-AmpX v2 and NGSgo-AmpX v3?
Both NGSgo-AmpX kits are designed as single locus amplification strategy featuring exactly the same coverage per locus. The difference between the two kits is the polymerase enzyme. NGSgo-AmpX v2 contains the GenDx-LongMix enzyme, and NGSgo-AmpX v3 uses GenDx-LongZyme. Both enzymes perform well, but GenDx-LongZyme (v3) shows a slightly more robust amplification results with les lot-to-lot variation.

How do I analyze the NGS data to get HLA typing results?
The FastQ files can be analyzed with the software package NGSengine. This is a dedicated software package for NGS HLA typing. NGS-Turbo and NGS-Pronto data is analyzed using NGSengine-Turbo.
How can we share data with GenDx?
Large data files (e.g. FastQ file) can be shared with GenDx through:
- WeTransfer (https://gendx.wetransfer.com/)
Never include personal identifiers of the data
Is it possible to use NGSengine or NGSengine-Turbo on a computer without internet?
If not connected to internet, copy the installer to the computer and use the ‘Activate offline’ option when following the on-screen instructions during installation. However, it is highly recommended to use ‘Online Activation’ to receive update notifications within the software.
