The field of transplantation diagnostics requires robust HLA typing to help diagnose HLA gene compatibility between donors and recipients. To support laboratories in achieving fast and reliable results, GenDx has published a comprehensive application note demonstrating the powerful capabilities of the NGSgo-ProntoAmp multiplex amplification assay. Whether your lab uses blood or buccal swab samples, NGSgo-ProntoAmp delivers a streamlined, high-throughput solution with excellent sequencing quality.

Proven performance in external clinical labs

For this study, three external clinical laboratories tested NGSgo-ProntoAmp using human clinical leftover samples. The laboratories successfully typed 80 human genomic DNA (gDNA) samples, isolated from both blood () and buccal swabs (). The samples were sequenced on both Illumina and Oxford Nanopore Technologies (ONT) platforms to ensure comprehensive compatibility.

Key highlights from the study:

  • Exceptional accuracy: The data analysis revealed highly accurate HLA typing results, showing 99,7% (Illumina) to 100% (ONT) concordance at the field resolution when compared to known pre-types. At the field resolution, all samples with available pre-types were 100% concordant.

  • High-quality data: Across both sample types and both sequencing technologies, the assay produced excellent sequencing data with balanced amplification and a clear discrimination between signal and noise.

  • Comprehensive coverage: The assay reliably amplifies 11 essential HLA genes (HLA-A, -B, -C, -DRB1, -DRB3, -DRB4, -DRB5, -DQA1, -DQB1, -DPA1, and -DPB1).

  • Rapid workflow: With three ready-to-aliquot mixes and a rapid PCR program of just 90 minutes, NGSgo-ProntoAmp offers a highly efficient solution for transplantation diagnostics.

Whether your lab prefers Illumina or ONT sequencing, NGSgo-ProntoAmp provides a high-throughput and high-resolution HLA typing solution you can trust.
