Is it possible to add an additional donor to an already typed recipient-donor combination?

To add additional Donor samples to an already existing Recipient-file you have to do the following:

1.    Select the existing recipient name from the dropdown-menu on the top left of KMRengine
2.    In the left-hand column the original sample and donor(s) will be displayed.
3.    In this same column select the orange  ‘plus ’ symbol marked ‘Add Donor’ to register details about the new donor.
4.    After filling in the details you can select KMRtype, only the additional Donor will be placed on the experimental plate.
5.    Type the additional donor with qPCR.
6.    Import the file from the qPCR machine into KMRengine.
7.    At this point KMRengine should recognise that the imported data belongs to a donor sample associated with your already existing recipient sample, and should call informative markers for the new recipient-donor combination.
8.    To use the Donor for a tracking experiment, select the recipient name from the dropdown-menu again, the donor data should be added to your tracking options.

Which qPCR platforms can be used with KMRengine?

Our multiplex chemistry will function with the emission filters of most standard qPCR platforms.
KMRengine is engineered to allow operators to choose with which machine they would like to use for their experiment.

Which channels of the qPCR system should be used for detection?

For KMRtype you need 4 channels:

  • FAM
  • JOE, VIC, HEX or Orange560
  • ROX or Red610
  • Cy5, Texas red or Quasar670 (passive reference)

For KMRtrack you need 2 channels:

  • FAM
  • Cy5, Texas red or Quasar670 (passive reference)

Which file formats can be analysed by KMRengine?

KMRengine loads files of the .txt and .csv formats.

What is the file type of the reports generated by KMRengine? Can they be exported to hospital LIMS?

The file types are PDFs and XML reports.

 

During plate-setup in KMRengine, is there a way to delete wells?

It is not possible to delete single wells, the undo button could be used multiple times consecutively to go back a number of steps.

When entering concentration in KMRengine during the experiment setup, is this the desired or actual concentration of the non-diluted sample?

This concentration is the initial undiluted concentration of the sample, the concentration given by you during setup is used to calculate the dilution step required in the automatically generated protocol.

What does “Date of transfer” refer to?

This is the date of transplantation.

Is it possible to manually enter informative markers in KMRengine?

Yes, it is possible to manually enter informative markers through one of the following two routes:

– Make a setup for a typing experiment including recipient and donor in KMRengine, perform the experiment and import the qPCR data into KMRengine. After approval of the data, return to the homescreen and select the recipient in the drop-down menu described above. The informative markers found in the typing will be displayed in the lower left screen. You can either select these markers for tracking by clicking on them or manually add additional markers using the button. Select the recipient in the ‘Reference Sample’ part of the screen and fill in details on your post sample. Then select KMRtrack on the top of the screen. Once you export this setup for tracking, the selected markers will be saved in your transplant file.

– If you already have typing data but have no KMRengine record of this, it is possible to manually set-up a tracking experiment without the use of imported KMRtype data. To do this, fill in the ‘Donor’ and ‘Recipient’ fields and select the recipient in the ‘Reference Sample’ field. Subsequently fill in the data for your post-transplant sample in the ‘Post-sample’ field. You can now manually add markers via the button. Once you export this setup for tracking, the selected markers will be saved in your transplant file.

The latest version is 2.7.0 released in May, 2023
